Sequence and where the open reading frame sits

Lee and colleagues, in Cell Metabolism in 2015, reported a short open reading frame in the mitochondrial 12S rRNA that encodes a 16-amino-acid peptide, and they named it MOTS-c, mitochondrial open reading frame of the 12S rRNA-c (PMID 25738459). The sequence recorded on this site's MOTS-c versus SS-31 comparison is Met-Arg-Trp-Gln-Glu-Met-Gly-Tyr-Ile-Phe-Tyr-Pro-Arg-Lys-Leu-Arg, one-letter code MRWQEMGYIFYPRKLR. Sixteen residues, an N-terminal methionine, and no disulfide in that written string are the identity marks this note uses. The glossary lines are MOTS-c, sequence, N-terminus, and linear peptide.

UniProt accession A0A0C5B5G6 lists that same 16-residue chain and a calculated mass of 2175 for the unmodified sequence, and it cites the 2015 paper (UniProt A0A0C5B5G6). That database mass is not the lot result. Copy the mass and the species from the file in hand. If they do not fit the 16-residue chain, stop and hold the vial. The 2015 paper also states that this frame is translated with the standard genetic code in the cytoplasm, because the mitochondrial code yields tandem start and stop codons. That is a biosynthetic description. It is not a preparation method.

What the 2015 cell model is for

Lee and colleagues describe cellular actions on the folate cycle and on de novo purine biosynthesis, with AMPK activation in the systems they measured. This note cites that class of model.
